The Expression of Dihydrodiol Dehydrogenase in Squamous Cell Carcinoma of the Skin

Squamous cell carcinoma(SCC) is one of the most common skin cancers and has many etiologic factors. Polycyclic aromatic hydrocarbons(PAHs) are one of the causes accounting for SCC. How PAHs react to human skin remains unclear. It is known that dihydrodiol dehydrogenase(DDH) is the major enzyme catabolising PAHs. Therefore, we addressed a question on the level of DDH expression in SCC. In this study, eleven cases of SCC were investigated immunohistochemically by DDH-specific polyclonal antibodies. Expression of DDH was further confirmed by in situ hybridization and immunoblotting. Our data showed: 1. ten of eleven SSC cases overexpressed DDH according to the results of immunohistochemistry, 2. five cases were confirmed for DDH expression by in situ hybridization, and 3. the only fresh SCC sample examined by immunoblotting was also positive for DDH expression. Our results indicate that the frequency of DDH expression is high in SCC.
